Inspired by a real-life story from Telangana, the film's story is set in a small village near Illandu. Raju Weds Rambai revolves around Raju, a young man who helps run his family's traditional band troupe. Raju falls in love with Rambai, played by Tejaswini, the daughter of Venkatanna (Chaitu Jonnalagadda), a compounder at a government hospital. Their budding romance faces obstacles from Rambai's father, who is more concerned about the social honour and status than the daughter's happiness. The film explores themes such as love, family expectations, social honour, personal sacrifice, and the impact of rigid social beliefs.
